Shimmy across to the other side of the room and open the gate, leading to a large room with lots of crates and a few enemies. When you land on a wooden platform above the water, face the ladder behind you and run up the wall next to it. This tomb can be found in Tuscany, right after Sequence 04-04. Use the shining window to leave the tomb and finish the memory. The top of the Duomo holds the sarcophagus you're after, which holds the Seal of Iltani. The rest of the climb will be pretty straight-forward. Don't jump from the chandelier towards the cross, but instead keep moving around the octagon to continue upwards. When you get to the next ladder, jump from the wooden platform you're on towards the octagonal frame to the east, and then jump from there towards a few more platforms to get to a chandelier. Keep traversing through the church by starting west of the ladder, then making your way east with more chandeliers and the sort. Jumping towards it will make the ladder fall, which sort-of serves as a checkpoint that you can use to get back up here if you fall. Continue climbing above the floor with the scaffoldings and chandeliers, and you'll eventually have to make a jump towards a wooden platform with a ladder.

Start by climbing up the door to the west with the small white piece on the ground. The entrance is at ground level on the northern side of the Duomo. You can get to this tomb in Florence as soon as you finish Sequence 04-04. You'll be able to leave the building by using a hidden passageway in the wall. After their conversation ends, take a brisk walk to the assassin sarcophagus at the end of the path and collect the Seal of Darius, the first of a collection of six. Climb up there and head inside to eavesdrop on Francesco's meeting with his fellow Templars.

Regardless of his survival, you'll end up in a circular room with a door fairly high up to the northeast. Kill him early if you can you can't use your throwing knives, unfortunately, but you'll still be able to air assassinate him if you have a high enough elevation over him.

Drop down on them and defeat the ones that are searching the room the last of the guards will flee for help upon seeing you, so you'll be chasing him all the way to the end of the catacombs (if he survives that long). Use it, and then keep climbing up to the send lever, which will open the door that the guards are supposed to be protecting. Look to the left to find a way upwards, where you'll find a very large wooden lever. Jump into the hay and enter the next room. By pulling this last switch, you'll give yourself access to a hay bale at the bottom of the room, and you'll also unlock a door leading further into the catacombs. After pulling the switch, jump across the opened path to the other side of the room and look north to find your next set of poles to hop on. You'll be sent into a rather tall spire, where you'll have to swing and jump your way to the next lever by using said objects to the left of where you came in from. Pull a switch on the right wall to open the gate ahead. Enter the catacombs with the skull in the wall, and you'll find yourself in a pretty small room.
